1980 xs650 spoke wheels

All years will fit with the correct spacers, but to make it easier use the later 80+ wheels. Those will be a bolt on deal if your hardtail maintains the factory spacing.
 
There were no rear disc spoke wheels used on the 650.

like james sez, they'll all fit, pick the wire wheels that you like the look of.
But
like 5twins sez XS650s never did have a rear disk wire wheel.
Your easy option is to switch to a drum brake rear.
Your difficult option is to DIY a disk rear by lacing a 16" rim onto a front hub and
following the "front wheel in rear" protocols you'll find by using the "search" button.

What kind of brake set up is it? I was looking at some spoke wheels on mikexs and wasn't sure which ones to get any guidance

Would these be a direct bolt on to my bike?

There is a hardtail that is being posted about by a new member at present and it has an XS650 front spoke wheel and hub with a disk brake on the rear.

Mikes sells a rear spoke disc hub but it’s not cheap. Using the front hub on the rear is also an option but also not cheap. If you wanna go rear disc most guys use a Harley wheel or some other model and make spacers etc to get it to work. Some skill level is required.
 
There are no bolt-ons, at least none that anybody has discovered yet.
 
A 76 CB750F had a rear spoked wheel with disc brake, while not a bolt on it could be fitted to you XS. The CB750 rear wheel is 18"
