This may be a dumb question or just may have a simple awnser, but here it is.
Why does the drive chain have a section where it is tighter than another section.
I always adjust my chain at the tightest run.Any thoughts?

Chains stretch unevenly so you end up with tight and loose spots.

Even a brand new chain has spots tighter than others.
They can't make every side plate exactly the same length or hardness. All the pins and rollers are slightly different too.
As the chain wears all these parts wear at different rates. Some links wear faster. This makes the chain longer in places. When these long spots are between the sprockets the chain is looser, when the short sections are between the sprockets, tighter.

You can have a tight and loose side if you parked on an incline. The drive sprocket stops turning but the rear wheel can turn until all the slack is gone.

You are correct in setting proper chain tension at the tight spot you should also do it with the normal operator weight on the seat comppressing the rear shocks
