Also, how did you pour it into the cylinders to begin with? I'm guessing a funnel thru the spark plug hole.

....did you put the plugs back in???
I have a friend that lives about 2 minutes away and he’s a mechanic.
I used a funnel to pour into the spark plug hole and I haven’t put the plugs back in yet.
Besides that one little moment the kick starter moved, I’ve been unable to move it since.
It feels like it’s stuck in gear, and I can’t get it out.
 
It it was me I would put the rear wheel back on if it spins ( Not locked Brake )
with chain.
No plugs in
Then try to spin the wheel and at the same time operate the gear shifter pedal having pressure on the pedal.
A bit back and forth on the wheel and if clutch handle moves try that also and on an XS 650 it shifts into and out of gear
with no clutch. Dont force it to hard
Ensure if it is in gear and if it can be moved in and out of gear and so

Stepping on the Kick and rear wheel spins it is in gear Not spinning in neutral

Being a 3 on a scale 10 I would advice against take to big steps at this point in time

Inspection of the tank comes soon so not dirt and rust gets sucked into the Carburetors

I was gifted a 1975 XS500 by a friend who just wanted it out of her garage, and I figured this was the perfect chance to start learning about motorcycle engines. After binge-watching hours of YouTube tutorials, the first thing I did was pour some Marvel Mystery Oil into the cylinders.

The first issue I ran into was that the rear wheel wouldn’t spin. I suspected the rear brake might be the culprit, so I loosened the rear tire enough to remove the chain—the tire spun freely, so that wasn't the issue. I’m about 93.2% sure the bike is in neutral. I also removed the clutch cable and sprayed some lubricant to help free it up, but it's still a tough squeeze.

Then I decided to put a little pressure on the kick starter just to see what would happen. Turns out, it sprays Mystery Oil all over the garage. So now I’m looking for advice on what the problem could be. I’m very new to this, so please explain things like I’m 12. Thanks!

Just to get everyone caught up, I was able to finally get the bike into neutral. Hooked it up to a battery charger turned the key on and saw a neutral light. Unhooked battery charger and applied pressure on the kick starter, I was able to get it about 3/4 down, but that’s it. How much pressure should I be putting on it? I don’t want to damage anything.
 
I believe that's a 180 degree engine. When piston 1 is at top dead center (TDC), piston 2 is at the bottom of its stroke.

So, does the kicker move smoothly then abruptly stop like you hit something? Make sure you don't have a valve stuck open.

That is good you are asking .So we can do it carefully

And in neutral --no more force than trying to step on the kick starter one foot still on the ground

I do have a little oil in the cyclinders
 
It can be removed by taking off the e-clips and tapping out the pins.

Take the plugs out and In gear can you roll the bike backwards (gently)?
Check Jetmechmartys' #31 post.
If you're getting partial movement my first concern would be interference
and not to cause any damage.
I'm not familiar with this specific engine,
but you definitely don't want to be smacking against a valve.

Thanks for the quick response. I wouldn't say I'm getting partial movement. It moved a little bit once, maybe twice. I've tried moving the bike back and forth while in 2nd & 3rd gears with no luck. Hopefully letting it soak a little longer will help out.
