Yes, that could cause issues. Those are vents for the float chambers. No venting might cause a vacuum to form in the bowl which could stop fuel flow.
I don't know why you guys insist on buying rebuild kits. It's well documented here that most are no good, containing many wrong parts. Those new kits you've ordered do not have the correct stock jet sizes but they may work for you if you've done mods and need larger than stock jets.
