XS 650 SPECIALS with wire wheels

If you are looking to do rear disk/spoke wheels you could use a Harley wheel.
This picture is 18" rear Harley wheel and Harley front and 21" wheel. I think most of the early
spoke bikes were all drum.

Did any of the Specials that came with wire wheels, have a 16 inch rear with a Disc brake, or were all the rear wire wheel drums?

AFAIK, all XS650 rear wire wheels had drum brakes.
If you really want a disk brake 16" rear wire wheel, try this:-
Get one of those Tracker kits that lets you fit a front wheel into an XS650's rear end.
Take the 19" rim off a front wire wheel and lace it's hub onto a 16" rim instead.
The stock XS650 rear disk, caliper, caliper mount and torque stay should work, possibly with some spacers and such to line things up correctly.
And note, if you don't have a stock XS650 rear disk, it's smaller than the front disk.
It's the same smaller disk that's on an XS750/850.

No, there were never any spoke disc rears of any size put on a 650 from the factory. As mentioned, all spoke rears were drum, either with a 16" or 18" rim.
